Okay, let's have some fun here speculating. I hope this plays out: teams travel all the way to Sudbury, all tired because of the KoR, go down deep into the mine, only to find Alex Trebek there with the next clue.

You're probably right, I am just having some fun.
But just to clarify, the KoR clue told them to go directly to the mines, and it's not part of the Detour. Nor is the ER/CPR task.

The Detour is the synchonized swimming, or a crime scene simulation that you see in the new preview on the website. The picture I found on Twitter shows a sign with the Detour options, "Analyze" or "Synchronize", and when I asked the poster for more information she said one was at the pool, and the other looked like a game of "Clue".

Also in that clip, by the way, Kristin apparently recovered during the flight because she's no longer on crutches. And Brian & Cynthia say in a confessional that they didn't tell anyone they're police officers. At the latter task, Brian says to Kristin "We don't know anything about policing. I hope we don't mess this up! (wink to the camera)"

We know the mines were first. No idea where the hospital task was, but it was presumably a Roadblock, since only Nic was shown doing it.
